MATRIARCHS BIOWORKS PRIVATE LIMITED is a groundbreaking, women-centric biotechnology startup based in North India, dedicated to making India self-reliant in healthcare. Our mission is to pioneer the biotechnologies that cater to the specific healthcare needs of our local communities, ushering in a biologic revolution that is accessible and affordable for the public.We developed CLing hybrid technology to produce multi-subunit protein complexes. We identified a pair of native E. coli proteins having high affinity (kD 10-17 M) for each other, and used them as fusion tags for heterologous protein subunits. We proposed that the affinity between two native protein tags could allow the subunits to come together in the periplasmic space, increasing the rate of folding and disulfide bond formation to make functional protein complexes. The fully folded functional protein complex can be eluted by Ni-NTA affinity chromatography followed by tag cleavage to provide biologically active therapeutics in high yields. We tested our system to produce Cimzia (Certolizumab), an anti-TNFα PEGylated Fab fragment for the treatment of auto-immune disorders such as rheumatoid arthritis. We achieved high and balanced yield of heavy and light chains of the Fab fragment, certolizumab, and now aim to purify the fully folded Fab’ fragment and develop it into our first product, named, Cimzelle, a biosimilar of biologic, Cimzia. We are developing Cimzelle as a biosimilar to an FDA-approved reference product, Cimzia, with the motive of reducing the product cost. Pfenex, USA has developed a Cimzia biosimilar PF688, which is currently undergoing preclinical/bioanalytical characterisation. Additionally, Biogen and Xbrane Biopharma have entered into a commercialization and license agreement for a Cimzia biosimilar candidate, Xcimzane. India solely depends upon the imports for meeting its growing demand of Cimzia (Certolizumab pegol).
