Brief Note on Innovativeness of product and services offered by Kanashi Biotech New Drug Discovery is the key area of Kanashi’s interest from the identification of novel targets for HIV, Tuberculosis (TB) and cancer in collaboration with eminent biologists and medicinal chemist to design and synthesis of novel scaffolds and New Chemical Entities (NCE’s) is the core competency of Kanashi’s discovery. To support our drug discovery program and financial viable model Kanashi offers Research and Development services in the area of ‘affordable medicines for all’ by taking R and D projects for novel process development for ‘Generic Medicines’ as well the Indigenous technology development for ‘Import Substitute’ key raw materials for the generic medicines at competitive rates with environmentally friendly / green process for the KRM’s. Our efforts in R and D are available to the industrial clients who do not have their own R and D team or scientific expertise to execute those programs, including units that are depending on import of key raw materials. Thus, Kanashi has contentedly helped in developing indigenous technology for products and services. In addition to this Kanashi also involve in novel process development of the specialty chemicals, the scaffolds and NCE’s used in early-stage drug discovery. At Kanashi these projects are executed by two ways one by collaborations with National and/ or International academic Institutes for synthesis of novel molecules on risk-sharing basis and secondly purchase order based research and custom manufacture of such molecules for initial discovery phase study at cost-effective way and help felicitate the drug discovery research keeping promise on delivering our tag line Discovering Innovations…… The uniqueness of Kanashi in taking R and D projects from NCE’s, Natural product API’s to speciality materials, generic API’s, Agrochemicals to basic raw materials; using the olden chemistry with new approach; using the frontier knowledge and methodologies in applying recent trends in chemistry to cost-effective, energy conservative and environmentally friendly solutions for manufacturing of these molecules at competitive and commercially feasible routes. The following examples of a few of the successfully delivered programs support our innovativeness in product and services. Early Stage Drug Discovery Programs 1) Established collaboration with Startup Biotech Company in Maryland, USA and successfully delivered one of the novel analog SP1 that is in Animal study for antidiuretic therapy. 2) Our collaboration with one of the prime university in the USA; Stony Brook University and Colorado State University on antitubercular (TB) infectious disease. Our results of this research had been published in well known Wiley journal Chem MedChem. Pan, P., Knudson, S. E., Bommineni, G. R., Li, H.-J., Lai, C.-T., Liu, N., Garcia-Diaz, M., Simmerling, C., Patil, S. S., Slayden, R. A. and Tonge, P. J. (2014), Time-Dependent Diaryl Ether Inhibitors of InhA: Structure-Activity Relationship Studies of Enzyme Inhibition, Antibacterial Activity, and in vivo Efficacy. ChemMedChem, 2014, 9: 776–791. 3) Our contribution in felicitating research work at IMTECH, Chandigarh and CSIR National Chemical Laboratory (NCL), Pune, By providing multistep small molecules on 10-100 gm quantity. 4) We have achieved a commercial process for the synthesis of biologically important DNA dyes Hoechst 33258 and 33342. Hoechst 33258 and 33342 are specific fluorescent DNA stains that bind within the minor groove of double-stranded AT-rich regions. These stains can be used on both live and fixed cells (1). Replicating or dividing chromosomes will not stain with Hoechst 33342 DNA dye, currently imported by most of the biology labs in India our efforts for “Lab to Commercialization” are on the way. 5) One of the key commercial processes for the manufacturing of Ethyl-3-(diethoxyphosphoryl)-4-oxopentonoate CAS NO 57648-56-3 on a Multikilogram scale has been successfully optimized and supplied to Lupin limited, India multiple times, however, our financial and restricted resources hampered our further scaling up of this molecule on a commercial scale. 6) Our Research and Development capabilities were successfully tested when we achieved “First total synthesis of N-[2-(5-amino-1,3,4-thiadiazol-2-yl)-4-fluoro-6-methylphenyl]-3-bromo-1-(3-chloropyridin-2-yl)-1H-pyrazole-5-carboxamide on 50 gm scale with purity > 98.8 % by HPLC in overall 10 steps in the record time of 45 weeks. This molecule has no literature evidence for synthesis and inventors were unable to complete synthesis. Affordable Medicines for All 1) One of the important classes of pharmaceuticals Non-steroidal Anti-inflammatory Drugs (NSAIDS) has a large market volume; Our efforts in addressing the commercially viable technologies with such big market will have a big impact on affordable medicines for all.
